Tetrahymena Thermophila, a single celled protozoa, contain several myosin proteins with mutated domains corresponding to similar mutations in human myosin which are linked to hereditary heart disease and hereditary deafness. This research attempted to create a recombinant vector plasmid for insertion into the protozoa, in order to express the Myosin 8 motor domain attached to a yellow fluorescent protein (eYFP). A reproducible protocol was created for the purification of Myosin 8 from gDNA and digestion of the vector plasmid. However, the process of ligation into a recombinant plasmid needs further investigation to complete reliably.
